ABSTRACT

Objective:To analyze and compare the differences between the epidemiological data and clinical indicators of confirmed and suspected undiagnosed cases of COVID-19 in Changning District, Shanghai. Methods:A retrospective comparative study was conducted. We included 20 confirmed and 34 suspected but undiagnosed COVID-19 cases from January 20 to February 29, 2020. We analyzed the differences in epidemiological history, early clinical symptoms, blood routine indicators, and clinical imaging characteristics between the two groups. Results:The epidemic status of COVID-19 in Changning District of Shanghai was mainly imported, and most cases were promptly confirmed. Early clinical symptoms of confirmed and suspected undiagnosed cases often manifested as respiratory symptoms such as fever and dry cough. Compared with the confirmed cases, the cell counts of leukocytes, eosinophils, and neutrophils in suspected undiagnosed cases were significantly higher. Also, the concentration of serum C-reactive protein in suspected cases was higher than that in confirmed cases (P=0.230). The clinical imaging manifestations of confirmed cases were mainly ground glass opacity (GGO) scattered in both lung leaves, while the suspected undiagnosed cases mainly manifested as plain patch opacity, and the distribution of GGO was more irregular. Conclusion:There exists significant difference in blood routine indicators and clinical imaging features between confirmed and suspected cases of COVID-19.

ABSTRACT

Objective: To study the relationship between primary tooth discoloration and caries in preschool children. Methods: 675 preschool children enrolled from 5 preschools in Suzhou City, China were examined for dental caries and tooth surface stain. Parents completed the questionnaires about their children's eating habit and living habit. The relationship between primary tooth discoloration and caries was analyzed with the respect to diet, hygiene, and the use of medicine or medical supplements. Results: Of the 675 children, 117(17. 3%; 52 boys and 65 girls) had tooth discoloration with no significant differences between gender(P> 0. 05). In the 117 children with tooth discoloration, 36(30. 8%) had dental caries, and in the 558 children without tooth discoloration 325 had caries (58. 2%, between the 2 groups, P< 0. 05). The incidence of discoloration was related with the frequency of consuming pigmented foods, vegetable and Chinese herbal medicines(P< 0. 05). The frequency of brushing teeth, use of water purifier and the water intake quantity were not significantly associated with discoloration of the teeth(P> 0. 05). Conclusion: The discoloration of primary teeth was negatively correlated with caries in primary teeth. Intake of pigmented food and medicine, vegetable and Chinese herbal medicine is related to tooth discoloration in preschool children.

ABSTRACT

OBJECTIVETo explore the mechanism of polypeptide extract from scorpion venom (PESV) on inhibiting angiogenesis.

METHODSThe H22 hepatoma tumor model was established by subcutaneously implanting H22 hepatoma cells into mice. The tumor-bearing mice were randomly divided into 4 groups, i.e., the control group, the high dose PESV group, the low dose PESV group, and the 5-fluorouracil (5-Fu) group, 10 mice in each group. The intervention was lasted for 14 days. The growth curve of the tumor volume was drawn and the inhibition rate calculated. Pathological changes of the tumors were observed by HE staining. The microvessel density (MVD) was detected using SP method. The protein expression levels of phosphatidylinositol 3-kinase (P13K), phosphoprotein kinase B (P-Akt), hypoxia-inducible factor-1 alpha (HIF-1 )alpha, and vascular endothelial growth factor-A (VEGF-A) were detected by immunohistochemical assay and Western blot.

RESULTSThe tumor inhibitory rate was 64.8%, 43.7%, and 32.4% in the 5-Fu group, the high dose PESV group, and the low dose PESV group. Compared with the control group, the protein expression of PI3K, P-Akt, HIF-1alpha, and VEGF-A were obviously inhibited by PESV and 5-Fu (P <0. 05,P <0. 01). The MVD also decreased in the high and low dose PESV groups (P < 0.05).

CONCLUSIONSPESV could inhibit the angiogenesis of H22 hepatoma. The mechanisms might be associated with suppressing the expression of PI3K, P-Akt, HIF-1 alpha, and VEGF-A.

ABSTRACT

Hepatitis B Virus [HBV] DNA polymerase transactivated protein 1 [HBVDNAPTP1] is a novel protein transactivated by HBV DNA polymerase, screened by suppression subtractive hybridization technique [GenBank accession no: AY450389]. The biological function of HBVDNAPTP1 was investigated in this study. We constructed a vector pcDNA3.1 [-]/myc-His A-HBVDNAPTP1 and used it to transfect acute monocytic leukemia cell line THP-1. HBVDNAPTP1 expression was detected by western blot analysis in the cells. A cDNA library of genes transactivated by HBVDNAPTP1 in THP-1 cells was made in pGEM-T Easy using suppression subtractive hybridization [SSH]. The cDNAs were sequenced and analyzed with BLAST search against the sequences in GenBank. Some sequences, such as CIP4, might be involved in apoptosis development. mRNA and protein expression of CIP4 was identified by Real time RT-PCR and western blot in THP-1 cells. HBVDNAPTP1 could down-regulate the expression of CIP4 at both transcription and translation levels. HBVDNAPTP1 may be involved in the positive regulation on the initiation of monocyte apoptosis. The result contribute to reveal the HBVDNAPTP1 biological functions and provide new evidences for further exploration of the regulatory mechanism of HBVDNAPTP1
